DEFINITION
Gellan Gum is a high-molecular-weight polysaccharide gum produced by a pure-culture fermentation of a carbohydrate with Pseudomonas elodea, purified by recovery with isopropyl alcohol, and then dried and milled. It is a heteropolysaccharide comprising a tetrasaccharide repeating unit of one rhamnose, one glucuronic acid, and two glucose units. The glucuronic acid is neutralized to mixed potassium, sodium, calcium, and magnesium salts. It may contain acyl (glyceryl and acetyl) groups as the O-glycosidically linked esters. It yields NLT 3.3% and NMT 6.8% of carbon dioxide, calculated on the dried basis.

IDENTIFICATION
A.
Sample solution: Hydrate 1 g of Gellan Gum in 99 mL of deionized water. Stir the mixture for 2 h, using a motorized stirrer and a propeller-type stirring blade.
Analysis: Draw a small amount of the solution obtained into a wide-bore pipet, and transfer it to a 10% calcium chloride solution.
Acceptance criteria: A tough, wormlike gel will form instantly.
B.
Sample solution: The remaining Sample solution from Identification test A
Analysis: Add 0.5 g of sodium chloride, heat the solution to 80C, stirring constantly, and hold at 80C for 1 min. Stop heating and stirring the solution, and allow it to cool to room temperature.
Acceptance criteria: A firm gel will form.

Alginates Assay:
Sample: 1.2 g of undried Gellan Gum
Analysis: Proceed as directed for the Procedure in the chapter.
Acceptance criteria: 3.3% to 6.8% of carbon dioxide on the dried basis
Lead: NMT 2.0 µg/g.
Arsenic: NMT 3.0 µg/g.
Limit of Isopropyl Alcohol: To pass the test by Chromatography. ---
Acceptance criteria: NMT 0.075%
Microbial Enumeration Tests and Tests for Specified Microorganisms: It meets the requirements of the tests for absence of Salmonella species and Escherichia coli. The total aerobic microbial count is NMT 10000 cfu/g, and the total combined molds and yeasts count is NMT 100 cfu/g.
Articles of Botanical Origin: 4.0% to 14.0% on the dried basis
Loss on Drying: Dry at 105C for 2.5 h. NMT 15.0%

DESCRIPTION
Gellan Gum occurs as an off-white powder. It is a highmolecular-weight polysaccharide gum produced by fermentation of a carbohydrate with a pure culture of Pseudomonas elodea, purified by recovery with isopropyl alcohol, dried, and milled. It is a heteropolysaccharide comprising a tetrasaccharide repeating unit of one rhamnose, one glucuronic acid, and two glucose units. REQUIREMENTS
Identification:
A. Prepare a 1% solution by dissolving 1 g of sample in 99 mL of deionized water. Using a motorized stirrer and a propeller-type stirring blade, stir the mixture for about 2 h. (Save part of this solution for Identification Test B). Draw a small amount of the solution into a wide-bore pipet, and transfer it into a solution of 10% calcium chloride. A tough, wormlike gel forms instantly.
B. Add 0.5 g of sodium chloride to the 1% deionized water solution prepared for Identification Test A, heat the solution to 80C, stirring constantly, and hold the temperature at 80C for 1 min. Stop heating and stirring the solution, and allow it to cool to room temperature. A firm gel forms.
Assay: A sample yields not less than 3.3% and not more than 6.8% of carbon dioxide (CO2), calculated on the dried basis.
Isopropyl Alcohol: Not more than 0.075%.
Lead: Not more than 2 mg/kg.
Loss on Drying: Not more than 15.0%.
